Summary : | The MDR phenotype seems to play an important role in the resistance of neoplastic cells to anticancer drugs. This phenotype is associated with overexpression of a 170 kDa membrane protein called P-glycoprotein (P-gp). This protein is considered to be a transporter wich pumps anti-tumor agents outside of the cells. P-gp is constitutively expressed in several normal tissues suggesting an important role in protection and/or detoxification. This protein is also frequently overexpressed in chemoresistant epithelial and hematological tumors. The discovery of the MDR phenotype led to the identification of agents wich are able to reverse this resistance. These molecules, selected on their capacity to inhibe P-gp function are, usually too toxic to be used in vivo . |
